چکیده

Pathways of signal transduction of red and blue lightdependent acidification by leaf epidermal cells were studied using epidermal strips of the Argenteum mutant of Pisum sativum. In these preparations the contribution of guard cells to the acidification is minimal. The hydroxypyridine nifedipine, a Ca -channel blocker, partly inhibited the response to both blue and red light, while the phenylalkylamine, verapamil, a Ca -channel blocker that has been shown in plant cells also to block K-channels, caused nearly complete inhibition. The Ca + -channel activator S(-)Bay K 8644 induced acidification when added in the dark and diminished the light-induced lowering of the extracellular pH. The Ca + -ionophores, ionomycin and A23187, also reduced the light response. Furthermore, the light-induced acidification was inhibited by the calmodulin antagonists W-7 and trifluoperazine, but not by W-5. These calmodulin inhibitors completely inhibited the red light-induced acidification, but inhibited the response to blue light by only 60-70%. In general, inhibition by compounds affecting Ca-calmodulin signalling was always stronger on the red light response than that on the blue light response (with the exception of verapamil that blocked both the red and blue light responses equally well). This differential effect on red and blue light-induced responses indicates a role for Ca + CaM signalling in both the red and blue light responses, while a second process, independent of Ca is activated by blue light.

Role of nitrogen content of pea (Pisum sativum L.) on pea aphid (Acyrthosiphon pisum Harris) establishment

The leaf nitrogen content is generally accepted as an indicator of food quality and as a factor affecting host selection by phytophagous insects. The alate pea aphids (Acyrthosiphon pisum Harris, Aphididae) were given a choice among non-nodulated pea plants (Pisum sativum L.) supplied with one of four nitrate-N levels (0, 3, 15 and 30 mM). When whole plants were exposed to aphids for 7 days, th...

A Comparison of the Effects of Chilling on Leaf Gas Exchange in Pea (Pisum sativum L.) and Cucumber (Cucumis sativus L.).

The effects of chilling on the photosynthesis of a chilling-resistant species, pea (Pisum sativum L. cv Alaska) and a chilling-sensitive species, cucumber (Cucumis sativus L. cv Ashley) were compared in order to determine the differences in the photosynthetic chilling sensitivity of these two species. For these experiments, plants were chilled (5 degrees C) for different lengths of time in the ...

Cellular and subcellular localization of endogenous nitric oxide in young and senescent pea plants.

The cellular and subcellular localization of endogenous nitric oxide (NO.) in leaves from young and senescent pea (Pisum sativum) plants was studied. Confocal laser scanning microscopy analysis of pea leaf sections with the fluorescent probe 4,5-diaminofluorescein diacetate revealed that endogenous NO. was mainly present in vascular tissues (xylem and phloem). Green fluorescence spots were also...

Is xylem sap calcium responsible for reducing stomatal conductance after soil

18 Understanding the regulation of calcium uptake, xylem transport and its impacts on growth and leaf 19 gas exchange is a subject that has received insufficient recent attention. Calcium (Ca) is unique 20 within the group of key elements required for plant growth in that it also has a role in cellular 21 signalling via regulation of changes in its cytoplasmic concentration. Its mobility, withi...

Inactivation of pea leaf phosphofructokinase by light and dithiothreitol.

When intact pea (Pisum sativum L.) plants are illuminated, the glycolytic enzyme phosphofructokinase is inactivated. In crude extracts the enzyme is inhibited by dithiothreitol. It would seem that this cytoplasmic enzyme, like glucose-6-P dehydrogenase, is light-inactivated when the enzymes of photosynthetic carbon metabolism are light-activated.
